Angiogenesis Inhibitors in Personalized Combination Regimens for the Treatment of Advanced Refractory Cancers

Background: Angiogenic factors are commonly activated in solid tumors and present a viable therapeutic target. However, anticancer treatment with angiogenesis inhibitors (AGI) is limited to few cancers, mostly as monotherapy not selected based on molecular indications. We aimed determine whether patient-specific combination regimens AGI other agents when multi-analyte tumor interrogation (ETA: Encyclopedic Tumor Analysis) can expand the scope of AGIs advanced refractory organ cancers improved responses. Methods: evaluated outcomes 60 patients advanced, who received ETA-guided targeted, endocrine or cytotoxic agents. Radiological evaluation response was followed by determination Objective Response Rate (ORR), Disease Control (DCR), Progression Free Survival (PFS) Overall (OS). Results: Among patients, Partial (PR) observed 28 cases (46.7%), Stable (SD) 29 (48.3%) (PD, within days) 3 (5.0%). The ORR 46.7% DCR 95.0%. At most recent follow-up median PFS (mPFS) 5.0 months OS (mOS) 8.9 months. There were no Grade 4 therapy related adverse events deaths. Conclusion: anti-neoplastic agents, yield over monotherapy.
